Once you get to the Tollhouse Basement, you can use the key to unlock the Vault door in BG3. This should then cause the “wall” to slide and reveal the hidden chamber behind it. In the basement, you can use Toll Collector’s key to open the door or use Knock Spell to open the door. As the three fake Paladins of Tyr will be present in the Toll House, things won’t go smoothly for you if you bring her along. If you do, you will have to fight the Paladins, so you should leave Karlach at Camp and then come to the Toll House to explore the basement there. Head into the house and open the Ornate Wooden Hatch to enter the basement. The main quest list for Act 2 is actually quite small and streamlined, but there’s a ton of content that pops up around that main quest. Your main objective from the end of Act I carries over here, as you’ll finally be able to access Moonrise Towers. They’ll often be somewhat near her, and the distant ones seem to get closer to her as the fight starts. So, just keep your party spread somewhat within the meter range of the boss to get the distant visages, while the other party members stay close to the boss and can hit the closer visages to her. Ideally, also have a tanky character be the one in close range of the boss, as she can deal some pretty high spell damage. So here you can place one heavy item (1) and (2) on each one of these stone chairs, e.g., by taking the large vases present on the table. Similarly, you can also use your party members to sit on each of these chairs as well. So once you spot a trap, you can place a box or barrel to block the poison gas vents.

If you have your party clumped up, then the fight could get slightly harder for you, especially on Tactician. So, spread out when possible, and try to keep your party at least 5-10 meters away from the next party member to avoid any splash effects safely. Pick up the key and then make your way back to the Toll House and then use the Ornate Wooden Hatch on the right side of the door.
You cannot pick this lock and you cannot damage it unless you can deal more than 22 damage per attack - likely a laborious process, at this stage. There is a key, however - climb down two ladders along the southern edge of the Toll House and follow the road downhill. Hold down the “alt” key as you go and you should find the Toll Collector’s Key along the rocks. This will open the door, and frankly, it’s the simplest way to get past the door under the Toll House. Use the Toll Collector’s Key and open the door to the east, but be wary, as there are vents on the ground.

I found the roots that lead to a secret cellar behind a bookshelf in the basement but, after exploring the cellar, I've found nothing down there. I can't imagine that there isn't a purpose for it and I was curious if anyone had figured out what that purpose is? Every time I search for BG3 Tollhouse on google it keeps directing me to the Act 1 Tollhouse with the pallies and the puzzle that involves sitting on the chairs. Even if I narrow the Search to include Reithwin I still get that puzzle answer lol.
If you don’t (perhaps you sneaked down here with the Reduce spell?) you can put some nearby vases on the chairs - their weight is apparently sufficient to trigger the door. Once the wall is lowered, be sure to disarm three more traps (two vents and a linear trap [Perception 10], [Sleight of Hand 10]). Failing that, there’s another way to get behind the door under the Toll House, but this takes some fairly specific circumstances. First, head around to the eastern end of the Toll House, where you can find a Burrow Hole [Perception 5], which is too small for you to enter, even if you’re a smaller race.

The interior is occupied by some paladins of Tyr - Anders, Tryna and Cyrel, the last of which is also a merchant, although she sells little of interest. If you talk to Anders you can start the aforementioned quest Hunt the Devil, but these three are otherwise uninteresting. Gerringothe Thorm can be found on the south wing of the top floor searching for gold. Reithwin Tollhouse is a location found in the south eastern area of Reithwin Town, directly south of the Mason's Guild. It will be the first location you reach if you arrive from across the central bridge south west of the Ruined Battlefield. Having your party spread around is certainly a strong strategy, largely thanks to the Visages doing various forms of AOE elemental damage to your party.
